What is 1V-LSD?
1V-LSD (1-Valeroyl-d-lysergic acid diethylamide), colloquially nicknamed “Valerie” in the research community, is a potent synthetic novel psychoactive substance (NPS) belonging to the lysergamide family. It emerged on the international chemical market as a legal alternative to classic LSD.
1. The Prodrug Mechanism & Pharmacology
The most critical feature of 1V-LSD is how it behaves biologically inside a living organism:
-
The Structural “Tag”: Chemically, 1V-LSD is identical to standard d-LSD, with the exception of a valeroyl group (C5 H9O, a 5-carbon chain) attached directly to the N-1 nitrogen atom of the indole ring system.
-
In Vivo Conversion: 1V-LSD acts as a prodrug. When ingested, metabolic serum and liver enzymes break down the molecule through hydrolysis, cleanly severing the valeroyl chain.
-
Receptor Affinity: Once the valeroyl attachment is cleaved away, the remaining liberated d-LSD targets the brain’s 5-HT2A serotonin receptors. This initiates the cascade of neural signaling that produces the compound’s intense psychedelic effects.
2. Why Doses Shift to 225 mcg
While older lysergamides like standard LSD or 1P-LSD are typically distributed in 100 mcg or 150 mcg units, 1V-LSD pellets and blotters are standardly manufactured at 225 mcg ($\mu\text{g}$). This shift is caused entirely by molecular weight:
-
Because a 5-carbon valeroyl chain is significantly larger and heavier than a propionyl chain or a bare hydrogen atom, 1V-LSD has a higher overall molecular mass (407.56 \text{ g/mol}) compared to normal LSD (323.43 \text{ g/mol}).
-
A large percentage of the weight in a 1V-LSD sample consists of the valeroyl group, which is discarded during metabolism. Therefore, 225 mcg of raw 1V-LSD is equimolar to (delivers the same amount of actual active LSD molecules as) a standard ~100–125 mcg dose of traditional d-LSD.
